On home soil, Mikitenko, 41, broke the Masters world record as she finished third while in the men's race, Kenyan Wilson Kipsang smashed the world record by a staggering 15 seconds.
Florence Kiplagat won the women's race in 2:21.13, ahead of fellow Kenyan Sharon Cherop in 2:22:28 followed by a delighted Mikitenko, third in 2:24:54.
Her time broke the record of 2:25.43 set by Russian Ludmilla Petrova: 2.25.43 in New York in 2008.
Mikitenko was all smiles as she reached the finished by the Brandenburg Gate, waving her right arm in celebration as she realised the time.
Kiplagat was always in control and Mikitenko moved into third as Sweden's Isabellah Anderson dropped out of the running around the 30km mark and from there the German charged to her record time.
Kipsang was outstanding as he won the men's race in 2:03:23, breaking the record, which Kenyan Patrick Makau set on this course two years ago.
It was the fifth time in a row - and the sixth in total - that Berlin had been the stage for the world record as Kipsang won from fellow Kenyans Eliud Kipchoge, who was second in 2:04:05, and Geoffrey Kipsang, third in 2:06:26.
